Comparison between conventional Western blots and the new CHIP approach.
CHIP has three clear advantages over conventional Western blotting
  • It uses far less primary antibody
  • It takes less time to achieve a result and the data is easy to interpret and visualize
  • It uses less membranes
Summary of results from western blotting experiment.
  • In unstimulated mast cells, there is little or no phosphorylation of the MHC on serine or threonine residues. After activation, the MHC becomes phosphorylated on both serine and threonine residues.
  • Both serine and threonine phosphorylation peak around 1-2 mins after activation, and then decreased, but the threonine phosphorylation remained for a longer period.
  • Using chemical printing technology to investigate protein phosphorylation with phosphospecific antibodies is much faster, requires fewer gels or blots and uses significantly less antibody when compared with conventional Western strategies.
  • Results obtained with the CHIP are very similar to those obtained using conventional Western blotting methods.
  • Where the sample can be limited or the source of the antibody is rare or expensive, the CHIP provides a powerful alternative to conventional Western blotting methods.
