- Following an integration of both proteomic and genomic approaches it was discovered that the majority of the proteins and their expression level was consistent with the gene expression level derived from cDNA microarray experiments.
- For example, from the microarray study we found that the KRT19 gene, a potential breast cancer biomarker, was 9 times over-expressed in MCF-7 than in CRL-2072. Its corresponding protein product CK-19 was also found to be over-expressed. This protein is either missing or 5-fold down-regulated in CRL-2072, with at least 7 isoforms.
- Further investigation on these isoforms might reveal the diagnostic value of this protein in cancer study.

Xcise was used to perform gel acquisition imaging, protein spot cutting and digestion, peptide extraction and deposition onto the MALDI target
plate. To each gel plug, 0.1µg trypsin (Promega Corporation) was used for digestion at 30 ºC overnight with shaking. After cleaning and
concentrating with m-C18 ZipTip (Millipore), the tryptic-peptide mixture was mixed with matrices (CHCA:DHB = 1:1, 10 mg/ml) and loaded onto a
384-well MALDI sample plate (Shimadzu).
